Survey of diphasic method for mass production of insect pathogenic fungi

The objective of the laboratory research was to produce two efficient and native strains belonging to Metarhizium anisopliae and Beauveria bassiana by using solid substrates (wheat, cooked rice, barley, sorghum and wheat bran) followed by liquid one (whey) at 25ºC. The results showed that maximum submerged conidia were significantly produced in the 50 ml of whey compared to dextrose agar broth media. Then, the solid substrates were inoculated by the submerged conidia gotten from whey. After 14 days, 1 g of each solid substrate was suspended in 100 ml of 0.02% Tween 80. The highest mean number of aerial conidia was obtained on rice (1.08×107 ± 2.42×105) and wheat bran (1.06×107 ± 1.86×105) without any significant difference for M. anisopliae. The percentage of germination was 100% for all solid substrates with the exception of sorghum (98.90%). For B. bassiana, wheat bran produced most conidia (5.14 × 106 ± 1.09 × 105). However, the handling of wheat seeds that produced 4.30 × 106 ± 3.71 × 104 was much easier than wheat bran for mass production of B. bassiana. Therefore, the rest of tests were carried out on wheat substrates. On barley substrate (39.20%), the lowest percentage of germination occurred. The number of conidia per 1 g of sieved powder for both fungi was 5.74 × 108 when were suspended in 50 ml of 0.02% Tween 80. In comparison to other studies, the present research clarified that the above described methods were efficient to produce the most amount of conidia.
