Essential oil of clove exposes fungicidal effect against Fusarium fujikouri, the causal agent of rice foot rot disease

Rice foot rot disease caused by Fusarium fujikuroi, is one of the most important seed-borne diseases in all rice-growing regions of the world. Seed disinfection is the most effective way to control this disease. Due to the overuse of chemical fungicides and their environmental hazards, it is necessary to achieve new and safe methods. In this study, firstly, the antifungal activity of four herbal plants clove, lavender, rosemary, and artemisia was assayed on Potato Dextrose Agar (PDA) medium containing the concentrations of 0, 100, 500 and 1000 ppm of their essential oils. The highest inhibition (100%) was observed in 500 ppm of clove essential oil (CEO). To obtain the optimum concentration with high inhibitory, the assay was repeated within PDA containing 0, 75, 100, 200, 300, and 400 ppm of CEO. Results proved the highest inhibition (100%) of mycelial growth at 300 ppm of CEO. In addition, its fungicidal effect was assayed and data revealed that concentrations of 300 and 400 ppm fully disturb the fungal survival. After 24 and 48h, the spore germination of F. fujikuroi was examined in water agar medium containing 0, 100, 200, 300 and 400 ppm of CEO in three replications. Results showed that at 24 and 48 hours, the germination of spores were prevented by 67% and 60% at concentration of 300 ppm, and 98 and 89% at concentration of 400 ppm, respectively. In summary, 300 ppm of CEO was chosen as the most optimal concentration with the highest antifungal effect for further studies.
