چکیده مقاله:
Meloidogyne spp., also known as root-knot nematodes, are considered a significant constraint in agriculture, orchards, and greenhouses. In this research, the impact of applying different amounts of natural mineral biochar from the Kouhbanan region (as a biological, active, stable, and organic compound with its genome sequence registered in the NCBI system) on the M. javanica was investigated in pistachio orchards in Kazemabad, Kerman. Natural biochar was applied to pilot orchard No. 1 at a rate of 15 tons per hectare and to pilot orchard No. 2 at a rate of 30 tons per hectare. In this study, nematode populations were evaluated after 6 months in soil treated with biochar and control soil in the pilot orchards by isolating second instar larvae from soil samples using the tray method. Molecular identification of the nematode species causing root-knot disease was carried out by using specific primers. Based on the molecular analysis, the isolated nematodes were identified as M. javanica species. The number of second instar larvae of Meloidogyne nematodes in 100 grams of soil in the control and biochar treatments in pilot orchard No. 1 was 43.33 ± 9.7 and 7.67 ± 2.52, respectively, and in pilot orchard No. 2 was 36.67 ± 7.64 and 0.0. The results showed that the use of 15 tons of natural biochar per hectare resulted in a 90% reduction in M. javanica nematodes, and the application of 30 tons of natural biochar per hectare in a pistachio orchard led to the death of all nematodes.
