Isolation and identification of Bacillus thuringiensis bacteria isolated from farm soil

چکیده مقاله:
Introduction: The use of microorganisms in agriculture can be a suitable alternative to chemical methods because they are more compatible with the environment. Bacillus thuringiensis is a beneficial bacterium that is toxic to some invertebrates such as insects and nematodes by producing large crystalline proteins during their sporulation process.
Materials and methods: 20 samples were randomly sampled from the soil depth of 5 to 15 cm around five healthy plant roots from the farms of Isfahan, Yazd, Fars, and Alborz provinces. Bacterial isolation was identified based on morphological characteristics, biochemical tests, and 16S rRNA sequencing. Also, the identification based on the Cry gene was checked and confirmed.
Findings: The soil samples of Isfahan province had the most strains of Bacillus thuringiensis bacteria. Also, based on biochemical tests including oxidation and reduction, lecithin hydrolysis, and squaline hydrolysis, the isolates were divided into two serotypes.
Conclusion: Based on the sequencing results, one of the isolated serotypes was the Bacillus thuringiensis strain, which was registered in the NCBI database as RTS-BT1 strain with accession number MN598646.1.
