Effect of native pseudomonad bacteria on wheat (Triticum aestivum L.) under drought stress condition

In this study, the growth pattern of 10 pseudomonad isolates from wheat rhizosphere in TSB supplemented with PEG6000 (-0.3, -0.6 and -0.9 MPa osmotic potentials) were evaluated, and the potential of these isolates were studied in vitro to produce IAA auxin, siderophore, ACC deaminase, hydrogen cyanide production, inorganic phosphate solubilization and improvement of wheat cv. Kavir seed germination parameters under different osmotic potentials. In the greenhouse, the effect of isolates was investigated on some morphological parameters i.e., percentage survival, length and biomass of wheat shoot and root at three watered levels, 100% FC, 50% FC, and 30% FC. UTP113 isolate had the all of PGP characteristics, and it had the most effect on the seed germination parameters, percentage survival and length and biomass of shoot and root. Effect of UTP113 isolate on chlorophyll (Chl) fluorescence of wheat leaves at 100% FC, 50% FC, and 30% FC was measured 14 days after application of drought stress. Comparison of mean values showed that UTP113 isolate increased Fv and Fv/Fm in severe drought stress. Therefore, it can be concluded that UTP113 isolate can tolerate relatively high levels of drought by making changes in some physiological characteristics with good efficacy. UTP113 isolate was characterized by partial 16S rRNA gene sequencing. This isolate clustered with P. fluorescens, based on 99% similarity. The sequence of UTP113 isolate was submitted in the NCBI GenBank database with accession no. OP117153.1. The phylogenetic tree was drawn by comparing this sequence with similar ones in the GenBank database by MEGA7.
