Assessing the Efficacy of some Bacillus species in controlling Meloidogyne javanica and Fusarium solani f. sp. cucurbitae in cucumber

In the initial screening, the biocontrol ability of fourteen Bacillus isolates against Meloidogyne javanica and Fusarium solani f. sp. cucurbitae were investigated through inhibition of egg hatching and larval mortality as well as dual culture test, production of antibiotics and volatiles. The role of metabolites secreted from selected Bacillus was evaluated in the process of attraction or repellence of nematode second instar juveniles (J2s), in reducing the gall index and increasing the growth factors of cucumber under greenhouse conditions. Two bacterial candidates including Bacillus sp.-Ba2 and B. pumilus-Ba8 prevented nematode egg hatching by 92.4% and 70.67%, respectively. Metabolites from bacterial suspension also led to 97.3% and 84.65% mortality in nematode J2s, while bacterial volatiles resulted in 90% and 83.35% mortality, respectively. Ba2 (84%) and B8 (59%) metabolites showed a high effectiveness in repelling nematode J2s in Petri conditions. In greenhouse experiments, both strains reduced the gall index from 5 to 3. In the double-layered pot, the positive effect of volatiles on increasing the growth factors of cucumber was visible compared with other isolates and uninoculated treatments. In the split root test, the number of galls and egg sacs in cucumber seedlings treated with Bacillus sp.-Ba2 was reduced by 37.83% and 39.17%, respectively. In the seedling treated with B. pumilus-Ba8, the two aforementioned studied factors showed a decrease of 46.62 and 51.54 %. Considering the soil-borne nature of the tested pathogens, studying the role of Bacillus volatile and non-volatile metabolites in the control of these group of plant pathogens is necessary.
