Biological control of Fusarium crown rot of wheat with Chaetomium species

چکیده مقاله:
Fusarium crown rot (FCR) is one of the most important fungal diseases of wheat that threatens wheat production worldwide. There are many studies with promising results on using Chaetomium spp. as potential antagonists against various plant pathogens. The current research was carried out to evaluate the efficacy of native isolates of Chaetomium species as a biocontrol agent against
F. pseudograminearum, one of the major causal agents of FCR in vitro. Forty Chaetomium isolates, recovered from wheat and barley in Northwest Iran, were evaluated by the dual-culture antagonism assays. The 90 mm Petri-dishes containing potato dextrose agar (PDA) were inoculated with 5 mm plugs from 7-day-old cultures of Chaetomium isolates on one side just 10 mm from the edge of the plate. After 2 days, plugs of F. pseudograminearum isolate IRAN 4686C were placed on the opposite side of the same PDA plates at a distance of 70 mm from those of Chaetomium isolates. Plates inoculated with F. pseudograminearum alone were used as control. All plates were incubated at 25°C with normal day light conditions. The experiment was repeated three times. The inhibitory effect was observed when the control Petri-dishes were covered with pathogen mycelium. Three isolates of Chaetomium fimeti including 54-3, 3-3 and 55-3-a showed greater inhibition to mycelial growth of F. pseudograminearum than other isolates, with inhibition rates of 38.82%, 37.29% and 35.76%, respectively. PCR amplification and sequencing of the β-tubulin (TUB2) gene confirmed that all these isolates belong to C. fimeti.
