Antagonistic effects of Bacillus thuringiensis against Xanthomonas translucens in wheat

Black chaff or bacterial leaf streak is one of the diseases of wheat causing damage to the phyllosphere of the plant. In this research, effects of wheat epiphyte bacteria in controlling Xanthomonas translucens and growth of wheat plants, were investigated. 100 isolates were tested by using of dual culture assay to identify antagonistic isolates. In total, 10 isolates which inhibited growth of the pathogenic bacterium in invitro condition, were selected for in planta biocontrol assay. The selected epiphytes were sprayed on wheat leaves and then the inoculated plants were kept in greenhouse. Experiments were performed in the form of a completely randomized design. Comparison of the lesions length in treated and non-treated plants showed that isolate Ph8 with 85.57±2.27% had the highest disease inhibition index. According to the results obtained from seed germination percent, seed vigor index and weight or length of wheat organs, Ph8 was selected as the best beneficial isolate. This isolate could produce cellulase, protease and HCN. The experiments were repeated three times. The Ph8 isolate could fix nitrogen, and produce Acc de aminase. Quantitative assessment of ammonia production as a nitrogen source for Ph8 was 25.9±1.49 mg/ml. IAA production and phosphate solubilization were also measured in this isolate. The Ph8 isolate was identified as Bacillus thuringiensis according to16S rRNA gene sequencing.
