Identification of saffron endophytic bacteria in Khorasan provinces with antagonistic activity against Macrophomina phaseolina

Saffron is introduced as the red gold in Iran. This crop is the most valuable agricultural product in Iran and the most expensive spice in the world. The presence of growth limiting factors and various pathogens that have reduced the production and quality of this valuable product, and also due to environmental problems in the use of various pesticides, the use of antagonistic bacteria, including endophytes would be useful for agriculture. Endophytic bacteria are non-pathogenic microorganisms of plants that can make their host resistant to pathogenic and non- pathogenic agents. In this study, in order to isolate endophytic bacteria, sampling was carried out from the leaves and healthy petals of saffron in Khorasan. 64 endophytic isolates were isolated from the collected samples. Dual culture assay showed, 17 isolates had antagonistic properties against Macrophomina phaseolina. Isolates K8 and G5 had the highest inhibitory hallo. The Saffron plants treated with these two isolates showed low disease symptoms on plants. The production of cellulase, chitinase, protease and lipase enzymes on antagonists was also investigated. The results showed that all isolates were able to produce lipase and protease. Also, 82.35% of antagonist isolates were able to produce cellulase and chitinase, which indicated the role of these enzymes in antagonistic properties. Sequence analysis of two selected isolates G5 and K8 showed that the isolates are similar to Pseudochrobactrum asaccharolyticum and Enterobacter cloacae, respectively.
