Efficacy of epiphytic bacteria to prevent citrus canker in greenhouse conditions

Citrus play an important role in wealth generation and economical development worldwide. Nevertheless, many diseases such as citrus canker cause considerable damage to this valuable crop. In this study, epiphytic bacteria were isolated from citrus trees in the northern regions of Iran and their antagonistic activity was assessed against Pantoea agglomerans identified as the causal agent of citrus canker previously, in vitro and in greenhouse conditions. A total of 311 epiphytic isolates were obtained from 100 plant samples containing leaves, stems, and fruit of various citrus trees (orange, lemon, lime, kumquat, and limequat) collected in Mazandaran and Gilan provinces. According to the results of hypersensitivity reaction on tobacco leaves and soft rot on potato slices, the antagonistic activities of 82 isolates were assessed against the pathogen using a Co-culture assay in which 36 percent isolates developed a growth inhibition zone. Also, the efficacy of 15 selected antagonistic isolates was tested on leaves of two-year-old Mexican lime tree with three replicates. The results showed that the effect of treatments in reducing the development of symptoms was significant at the statistical probability level of 1%. The isolate 22P3 reduced the development of symptoms by 9.84% compared to positive control. Based on the phenotypic characteristics, the effective isolates in the laboratory and greenhouse conditions belonged to the genus Pseudomonas.
