Identification of some susceptible tomato genotypes to tomato yellow leaf curl virus in south of Kerman province

Tomato yellow leaf curl virus (TYLCV) belongs to the genus Begomovirus and family Geminiviridae. TYLCV infection induces severe symptoms on tomato plants and causes serious yield losses worldwide. TYLCV is persistently transmitted by the whitefly, Bemisia tabaci. To identify susceptible tomato genotypes to the Iranian isolate of TYLCV (G4) in some cultivated genotypes in south of Kerman province, the seeds of 7 tomato genotypes were prepared and cultivated in separated pots in controlled condition in greenhouse. Agro-inoculation of TYLCV were done using constructed infectious clone of TYLCV (from previous research) on every genotype (3-4 leaf stage). After four weeks, virus symptoms were surveyed. All cultivated tomato genotypes had demonstrated various degrees of disease symptoms. Based on severity of symptoms, Matin, Eden and Cherry tomato genotypes were identified as disease-susceptible cultivars among the seven studied genotypes. To confirm infection of TYLCV in symptomatic inoculated plants, total DNA was extracted from leaf and PCR was performed using TYLCV-1073-F/TYLCV-2046-R specific primers. Mild symptoms were observed in the other studied genotypes 3 month after inoculation. Disease severity in susceptible genotypes was graded from 0 to 4 based on the severity of symptoms on plants and cultivars were divided into three groups according to the intensity of the response. Therefore, it seems that susceptibility of tomato genotypes in south of Kerman province is different against TYLCV. Identification of resistant genotypes and recommendation for planting has an important key role in the integrated management of tomato leaf curl disease.
