ACC Deaminase, an important enzyme in Agaricus bisporous growth promotion

Some strains of benefical bacteria contain ACC deaminase enzyme. This enzyme converts ACC compound (a valuable Nitrogen source for bacteria) into ammonium and -α-Kectobutyrate. This enzyme plays an important role in facilitating growth by reducing the amount of ethylene (which is one of the spoilage factors in edible mushrooms), especially in conditions of environmental stress. Therefore, the aim of this study is to express, purify and determine the optimal conditions for activity of ACC deaminase (ACCD) enzyme in B. velezensis strain. After the investigations carried out regarding the presence of ACCD enzyme in B. velezensis, the optimal conditions of enzyme gene expression were investigated in the presence of different treatments. The results showed that by optimizing the gene expression of this enzyme in B. velezensis bacteria, the mycelium growth rate increased by 20% and the yield increased by 28% and the amount of ethylene decreased by 20%. Therefore, it can be concluded that the expressed bacterial ACCD gene degrades ACC and reduces ethylene synthesis and eliminates the inhibitory effect of ethylene on mycelium growth and primordium formation in A. bisporus. In this way, it helps to increase growth and productivity.
