Effect of culture media on growth and biocontrol efficacy of Trichoderma

In this study, the effect of four culture media including First) Mol.C.N.G. (50ml of sugar beet molasses, 5ml of corn steep liquor, 25g of NaCl and 25g of glucose), Second) Mol.C.G. (50ml of sugar beet molasses, 5ml of corn steep liquor and 25g of glucose), Third) Mol.C. (50ml of sugar beet molasses and 5ml of corn steep liquor), and Fourth) Mol. (50ml of sugar beet molasses) with a pH 5.5 were investigated on the growth and antagonistic efficacy of Trichoderma atroviride Tr9 against Phytophthora drechsleri, the causal agent of cucumber root and crown rot. One ml of suspension containing 108 spores of antagonist was inoculated at culture media and incubated at 130 rpm at 28°C for 14 days. Maximum sporulation (1.9×1010 spore/gdw) was observed in Mol.C. in laboratory studies, all treatments prevented the growth of the pathogen in different rate, but the Mol.C. with 56.33% showed the greatest effect on inhibiting of the pathogen growth, and in terms of colonization, Mol.C. and Mol. showed the greatest impact with 15.67% and 14.5%. Also, the highest rate of antagonistic radial growth was observed with 51% in biomass obtained from Mol.C.N.G. and Mol.C. Under greenhouse condition, biomass of Mol.C.G., Mol., Mol.C.N.G. and Mol.C. were able to reduce disease severity by 53.11%, 50 %, 50.33% and 50.11%, respectively. The biomass obtained from Mol.C. also showed the greatest effect on increasing shoot length, root length, shoot fresh weight and root fresh weight by 106%, 100%, 148% and 75%, respectively.
